摘  要:目的:探讨垂体大腺瘤中PTTG和FGF 2的表达与肿瘤生物学行为的关系。方法:用免疫组化技术(LSAB法)分别检测垂体大腺瘤(4 0例)和垂体微腺瘤(11例)中PTTG和FGF 2的表达。结合临床影像分级资料,分析PTTG和FGF 2的表达与大腺瘤生物学行为的关系及临床意义。结果:40例垂体大腺瘤中均发现PTTG和FGF 2的表达增高,两者的表达水平均高于其在垂体微腺瘤中的表达(t=3 .845 ,P <0 .0 1;t =4.3 81,P <0 .0 1)。PTTG和FGF 2在垂体大腺瘤中的表达水平显著相关(r =0 .64 0 ,P <0 .0 1)。PTTG的表达与大腺瘤侵袭性生长的程度显著相关(P <0 .0 5 )。结论:PTTG上调FGF 2的表达机制在促进垂体大腺瘤的生长过程中发挥了重要的作用,并有可能为临床预后的评估提供可靠的依据。Aim:To investigate expression of PTTG and FGF-2 in pituitary marcroadenomas and analyze relationships between gene expression and clinical tumor behavior. Methods:40 cases of pituitary macroadenomas operated from Jan.2001 to Dec.2003 were analyzed for PTTG and FGF-2 protein expression by immunohistochemical method. 11 cases of pituitary microadenomas were used as the control group. Clinical data including radiological classification were used to determine any associations between PTTG expression and clinical tumors behavior. Results:Expression of PTTG and FGF-2 were increased significantly in macroadenomas compared with pituitary microadenomas ( t =3.845, P <0.01; t =4.381, P <0.01). A significant correlation was found between PTTG expression and FGF-2( r =0.640, P <0.01). Furthermore, significant correlations of PTTG expression were found in tumors that invaded cavernous sinus and suprasellar extension ( P <0.05). Conclusion:Our results have supported the mechanism for PTTG-mediated up-regulation of FGF-2 as a fundamental role in promoting growth of pituitary tumors. Expression of PTTG may be related to cavernous invasiveness of pituitary macroadenomas. Thus, as the clinical marker of tumor behavior, PTTG may be used for evaluation of clinical prognosis.
