摘  要:目的:阐明NO对大鼠生精能力的影响及其作用机制。方法:大鼠去垂体,观察硝普钠(SNP)与N-ω单硝基L-精氨酸甲酯(L-NAME)对大鼠生精功能的直接作用。用放射免疫法测定血清内卵泡刺激素(FSH)、黄体生成素(LH)及睾酮含量;Greiss法测定血清NO-x含量;流式细胞术测定各生精细胞DNA含量,计算1c、2c及4c期细胞的百分比。结果:各组血清LH、FSH和睾酮浓度显著下降或逐渐下降。SNP组血清内NO-x含量显著升高,L-NAME组血清内NO-x含量显著降低;对照组、L-NAME组和SNP+LNAME混合注射组的细胞分布均以1c为主;而SNP组的细胞以2c为主。结论:体内高浓度的糖皮质激素可屏蔽LH和FSH的分泌,进一步抑制睾酮的分泌;SNP与LNAME可通过NO直接影响生精细胞的凋亡。Objective:To evaluate the effect of nitric oxide (NO) on spermatogenesis in rats and to explore its modulating mechanism. Methods: The rat model of pharmacological hypophysis shut off was built. The concentration of serum LH, FSH and testosterone were measured by radioimmunoassay.Serum NO-_x (nitrite/nitrate)concentration was measured by Greiss method. DNA of spermatogenic cells was detected by flow cyctometry (FCM), and the percentages of 1c, 2c and 4c germ cells were calculated. Results: Plasma levels of LH, FSH and testosterone (T) of every group declined significantly or gradually. In sodium nitroprusside (SNP) treatment group, the concentration of NO-_x was significantly higher in comparison with normal saline subjects; the number of 2c cells was higher than that of 1c and 4c. The concentration of NO-_x was significantly lower after Nω-nitro-L-arginine methel ester (L-NAME) treatment compared with normal saline subjects. However, in rats treated with NS and L-NAME, the number of 1c cells was higher in comparison with both 2c and 4c. No changes was observed in SNP+L-NAME group. Conclusion: The model of hypophysis shut off can be built up with high concentration dexamethasone in vivo, withdrawing the secretion of LH and FSH rapidly and inhibiting the secretion of testosterone (T).There is direct effect on apoptosis of germ cells in rats treated with SNP and L-NAME.
