摘 要:在分析antiHAVIgGCHO细胞对筛选的培养基中氨基酸利用的基础上,对基础培养基进行初步优化。再采用统计学正交分析方法,在初步优化的基础培养基中添加胰岛素、转铁蛋白等促细胞生长因子,研制了一种适于分泌抗甲肝病毒基因工程单抗的rCHO细胞悬浮生长用无血清培养基,经Spinner转瓶实验,细胞生长和抗体表达与进口同类产品相当,且成本低廉,可用于大规模悬浮培养。Based on the result of analysis of the utility ratio of amino acid in selected media for anti HAV IgG CHO cell,the media was improved by adding the relevant amino acids. The optimized media, by means of a statistical optimization approach, was supplemented with some cell growth factors, such as insulin, transferrin etc. Then a serum-free media for anti HAV IgG CHO cell was found. The result of Spinner bottle experiments proved that the growth rate and expression level of antibody in it were as the same as in imported products. It is much cheaper than the imported products, so it can be used in large-scale suspended batch cell culture.
