高效液相色谱法测定贯叶连翘中金丝桃甙的含量

摘　　要：目的:采用高效液相色谱法测定贯叶连翘中金丝桃甙的含量。方法:色谱柱:YWG-C18;流动相:乙腈-磷酸盐缓冲液(70:30,v/v),流速:1.0m1.min-1,检测波长370nm。结果:金丝桃甙的线性范围为0.0440～0.4398μg,r=0.9998(n=5),平均回收率98.5%,(RSD=1.4%n=5),方法精密度为(RSD=1.1%n=5)。结论:本法具有准确性、灵敏性、专一性,为测定中药贯叶连翘中金丝桃甙的含量提供了新的方法。Objective: To determinate hyperin in hypericum perforatum L.by high performance liquid chromatography ( HPLC ). Methods:An HPLC method was carried out by using an YWG-C 18 column, the mobile phase consisting of acetonitrile-disodium hydrogen phosphate(70:30,v/v) and the flow rate of 1.0 ml.min -1.The ultraviolet detection was at 270nm.Results:The linear range of hyperin was 0.0440～0.4398μg with a correlation coefficient of 0.9998.The average recovery was 98.5%,(RSD= 1.4%n=5)and the precision of the method was 1.1%.Conclusion: This method is simple, specific and accurate.
