银杏叶黄酮提取工艺及对Hela细胞Bcl-2 mRNA表达的影响  被引量:9

摘  要:目的银杏叶黄酮提取工艺优化及抗肿瘤作用的研究。方法采用碱液法、乙醇法、丙酮法3种不同的提取方法,设定不同的固液比、提取时间,优化出最佳条件,用于银杏叶黄酮类化合物的提取。无血清培养Hela细胞,以银杏叶黄酮提取物为诱导剂,24h后用RTPCR方法检测Hela细胞中癌基因Bcl2的表达情况。结果70%乙醇、固液比1∶3、回流时间为2h的条件下,提取银杏叶黄酮类化合物得率最高。采用RTPCR方法检测显示,银杏叶黄酮类化合物作用Hela细胞24h后,Bcl2mRNA表达水平下降。结论70%乙醇、固液比1∶3、回流时间为2.5h,为银杏叶黄酮提取最佳条件。银杏叶黄酮能下调癌基因Bcl2mRNA表达水平,从而可能有抗肿瘤作用。Objective To study the extraction technology optimization of flavonoids of Ginkgo bilobal and the anti tumour of the extraction of Ginkgo bilobal(GBE).Methods Different methods of alkali,alcohol and acetone were used.The ratio of solid to liquid and extrating time were optimized.Hela cell was cultured in RPIM 1640 without calf and in GBE as revulsant.After 24h,the expression of Bcl 2 mRNA was measured.Results The result showed that the best process was that alcohol concentration was 70%,the ratio of solid to liquid 1∶3,and reflux for 2h.After 24h,the expression of Bcl 2 mRNA by RT PCR were found decreased by GBE.Conclusion The best process is that the concentration of alcohol is 70%,the ratio of solid to liquid 1∶3,and reflux for 2h.GBE can decrease the expression of Bcl 2 mRNA.And it may has anti tumour effect.
