摘 要:目的:建立白果内酯的配位色谱分离纯化方法。方法:10g 白果内酯初提物经柱前与0.01mol·L^(-1)配位剂-硼砂反应,0.1 mol·L^(-1)盐酸调 pH 到5.0左右,常压硅胶柱层析,石油醚-乙酸乙酯混合溶剂梯度洗脱。结果:Na_2B_4O_7配位色谱法所得白果内酯的纯度为82.1%和76.9%;回收率为75.5%和72.1%。较普通色谱法纯度提高7.4%-12.6%,回收率提高11.9%-15.3%。结论:本方法操作简便,产品纯度和得率较高,可用于白果内酯的分离纯化。Objective:To develop a method for the isolation and purification of bilobalide according to the principle of coordination chromatography.Method:The bilobalide Extract(10g)was treated with 0.01 mol·L^(-1) sodium bo- rate.The mixture was adjusted to pH 5.0 by 0.1 mol·L^(-1) hydrochloric acid.After absorption of the bilobalide Extract onto silica gel,elution was carried out with a gradient elution of the ratio of ethyl acetate to petroleum ether. Results:The purities of bilobalide separated by borate coordination chromatography were 82.1% and 76.9%,which were 12.6% and 7.4% higher than the blank column chromatography,recoveries were 75.5% and 72.1%,which were 15.3% and 11.9% higher than the blank column chromatography.Conclusion:The developed method is sim- ple,reproducible and easy to operate.
