摘 要:利用人单核细胞表面有纤维连接蛋白受体的特点,我们建立了一种分离、纯化人外周血单核细胞的方法。该法制备的细胞悬液单核细胞占97%以上,纯度明显优于Ficoll-Hypaque密度梯度离心法。单核细胞获得率近5倍于传统粘附法,且细胞活性保持在97%以上,是一种分离人外周血单核细胞的简单而有效的方法。simple and efficient method of monocyte separation was developed based on the fact that fibronectin receptors are present on the membrane of human monocytes. The percent of monocytes was beyond 97% in cells obtained by this method , it was significantly high than that obtqined by Ficoll-Hypaque centrifugation. The yield of monocytes was 5 times more than that with classical adherance method , and the cell viability was always excellent (> 97% ) . We concluded that this method is a simple and useful technique in separating human peripheral blood monocytes.
