异种骨移植中多种免疫因子的表达及转化生长因子-β的调节作用

摘　　要：目的:观察异种骨移植局部白细胞介素-1(IL-1)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)mRNA和蛋白质的表达,探索转化生长因子β(TGF-β)对这些免疫因子的调节作用。方法:将复合了TGF-β的牛松质骨骨粒或单纯牛松质骨骨粒植入小鼠股部肌肉内,于植入后第4,7,14及21天取材检测。结果:在异种骨移植标本中多种细胞成份可表达I-L1α、IL-6及TNF-αmRNA和蛋白质。复合TGFβ的骨移植局部IL1-α、IL-6和TNF-αmRNA的表达明显低于单纯异种骨移植(P<0.01)。结论:在异种骨移植局部,多种细胞可以表达IL-1α、IL-6及TNF-αmRNA和蛋白质,且这种表达受到TGF-β的调节。Objective: To observe the expression of interleukin (IL)-1α, IL-6 and tumor necrosis factor (TNF)-α mRNA and proteins in local area of xenogenesis bone transplantation, and investigate the modulatory effect of transforming growth factor (TGF)-β on these immune factors. Methods: The bovine cancellous bone granules, which combined TGF-β or not, were implanted into the thigh muscles of mice. The samples were taken 4, 7, 14 or 21 days after implantation. Results: A variety of cells could express IL-1α, IL-6 and TNF-α mRNA and proteins in all specimens of implantation. The expression of IL-1α, IL-6 and TNF-α mRNA in the tissue around bone granules combined TGF-β was much lower than that in cancellous bone alone (P<0.01). Conclusion: In the region of bone xenograft, a variety of cells express IL-1α, IL-6 and TNF-α mRNA and proteins, and their expression is regulated by TGF-β.
