摘 要:报道不同溶剂体系以及工艺条件下的再生蜘蛛丝蛋白溶液以及丝蛋白膜的分子结构特征。以大腹圆蛛牵引丝为研究对象,分别用六氟异丙醇(HFIP)和溴化锂(LiBr)为溶剂制备再生蜘蛛丝蛋白溶液和丝蛋白膜。采用SDS凝胶电泳法测定了再生蜘蛛丝溶液的分子量。用拉曼光谱分析了蜘蛛丝蛋白溶液和丝蛋白膜的分子构象。研究了温度对再生蜘蛛丝蛋白膜和蜘蛛腺体蛋白膜分子结构的影响,并用红外光谱对HFIP溶解的再生丝蛋白膜的分子构象进行了分析。The spinning solution is one of the most important factors in the spinning of regenerated spider silk. This investigation described the comparative structural characters of regenerated spider dragline silk solution obtained in different solvents and technologies. The results of SDS gel electrophoresis showed the silk dissolved in LiBr had wider molecular weight distribution than in HFIP. The secondary structures of the silk film cast from the LiBr solution were α-helix,(β-sheet) and random coil. But the film obtained from HFIP solution was dominated by α-helix and random coil. Raman spectra showed the conformation of spider silk film dried from LiBr regenerated spider silk solution didn't have obvious change with temperature, otherwise, the conformation of a natural major ampullate gland protein film changed dramatically from predominantly α-helix at 30℃ to principally β-sheet at 60℃.
