五灵胶囊抗肝细胞损伤的药效成分分析

摘　　要：目的:研究五灵胶囊抗肝细胞损伤的药效成分及保护作用。方法:采用高效液相法(HPLC),将已知的对照品作对照,测定五灵胶囊中所含相应成分的含量;以体外D GlaN、LPS诱导的大鼠原代肝细胞为靶细胞,经细胞培养观察五灵胶囊及对照品对肝细胞损伤后的保护作用。结果:在与对照品对照条件下,经HPLC分析五灵胶囊中各成分的滞留时间与各对照品时间一致;五灵胶囊和其他3种对照品可使D GlaN、LPS诱导损伤原代肝细胞的ALT、LDH L、MAO、GSH ST、MDA水平显著降低,有效地拮抗D GlaN和LPS对肝细胞的损伤;在受损肝细胞内测定五灵胶囊的化学成分有五味子醇甲、五味子乙素、隐丹参酮、丹参酮ⅡA。结论:HPLC分析和细胞药效学证实,初步推断五灵胶囊抗肝细胞损伤的部分有效成分是乙素、隐丹参酮、丹参酮ⅡA,而且这些成分对肝细胞的损伤具有一定的保护作用。AIM: To investigate the chemical components and protective effect of Wuling Capsule. METHODS: With the pure monomer component as control, we analyzed the components of Wuling Capsule by HPLC. Hepatic cell injure model was prepared by the induction of D GlaN and LPS in vitro separately, and then compared the two different protective effects after liver cell culture. RESULTS: Both Wuling Capsule and pure component could obviously depress the levels of ALT、LDH L、MAO、GSH ST which were higher in damaged primary hepatic cells induced by D galN and LPS. The Schisandrin B、Crytotanshinone、Tanshinone Ⅱ A were found in hepatic cells. CONCLUSION: It is suggested that Schisandin B、Crytotanshinone、Tanshinone Ⅱ A were effective chemical components of Wuling Capsule against liver cell injure.
