摘 要:目的研究黄芪丹参复方成分提高胎盘血供的分子机制,为临床有效防治胎盘血供不足所致妊娠并发症提供思路。方法提取分离黄芪、丹参复方成分,运用NOS(一氧化氮合酶)阻滞剂L-精氨酸甲酯(L-NAME)制作一氧化氮合成阻滞大鼠模型,采用原位杂交(ISH)法观察胎盘滋养细胞诱导型一氧化氮合酶(iNOS)、单核细胞趋化蛋白-1(MCP-1)mRNA表达变化及黄芪丹参复方有效成分的影响作用。结果一氧化氮合成阻滞模型组滋养细胞MCP-1mRNA表达染色明显增强,空白组、黄芪丹参复方成分治疗组及硝酸甘油对照治疗组与模型组比较差异均有显著意义(P<0.01)。模型组iNOSmRNA表达明显增加,与黄芪丹参治疗组、阳性药物硝酸甘油组均有显著差异(P<0.01)。结论黄芪丹参复方成分降低母胎界面妊娠免疫反应,阻抑胎盘免疫病理损伤,可能与抑制iNOS、MCP-1mRNA表达及母胎界面炎性细胞游走、浸润有关,是中药能够提高胎盘血液供应的机制之一。the effective components of Mongolian milkvetch root -Danshen root, apply L-Arginine (L-NAME) to pregnant rats to make Nitrogen Oxide Synthesis blocking model. Observe the changes of the expression of iNOS mRNA and MCP-1 mRNA of placental nourish cells and the effection of the components of Mongolian milkvetch root -Danshen root compound on those expressions by the method of in situ hybridization. Results The expression of nourish cells MCP-1 mRNA of model group was obviously dyed deeper. The expression of blank group, treat group, treat group by the components of Mongolian milkvetch root -Danshen root compound and contrast treat group by nitroglycerin were significantly different from that of model group (P <0.01). The expression of iNOS mRNA of model group was obviously higher, and significantly different from those of treat group by Mongolian milkvetch root -Danshen root and treat group by positive medicine of nitroglycerin (P <0.01). Conclusion The components of Mongolian milkvetch root -Danshen root compound can restrain the immune reaction of placenta and restrain the placental immune pathologic damnification, which may relate to its function of preventing the expression of iNOS mRNA and MCP-1 mRNA and preventing inflammative cells roving and invading. Such of above is one Chinese medicine mechanism of elevating placental blood supply.
