摘  要:建立了尿样中MDMA及其代谢物MDA的毛细管电泳高频电导法检测分析方法。对电泳分离缓冲介质的种类、浓度、pH值以及分离操作电压和进样时间等实验条件进行了优化。缓冲液为2.5mmol LNH4Ac+0.5mmol LHAc+2mmol LSDS+10mmol LβCD,分离电压为15kV时,可实现较好的分离与检测。该法在2 5~80μg mL范围内,MDMA和MDA线性相关系数r分别为0 998和0 999,检出限均为1 0μg mL(S N=3)。不同添加浓度水平尿样,日间和日内RSD均小于5%,回收率均在90%以上,适于法医毒物分析和临床药物监测的需要。A method has been built for the determination of MDMA and its metabolite in urine by capillary electrophoresis with high frequency conductivity detection (contactless conductivity detection). With optimized buffer of 2.5 mmol/L NH_4Ac+0.5 mmol/L HAc+2 mmol/L SDS+10 mmol/L β-CD, the samples were well separated and detected at a voltage of 15 kV. The assay exhibited a linear range of 2.5~80 μg/mL for both MDMA(r=0.998) and MDA (r=0.999), and the limit of detection (LOD) of 2.5 μg/mL (S/N=3) for both. The intra- and inter-day coefficients of variation were less than 5% and the recoveries were better than 90% at various spiked levels. The validated method fully meets the requirement for MDMA and MDA analysis in urine in forensic medicine and clinical drug monitoring due to its simplicity,rapidity, and sensitivity.
