摘  要:目的比较家兔灌服桃核承气汤和单味大黄后大黄酸的药代动力学特征。方法家兔灌服桃核承气汤5.0g生药/kg和单味大黄1.4g生药/kg后,血浆经高氯酸和乙醚萃取,用高效液相色谱法测定大黄酸血药浓度。色谱柱为EclipseXDBC18柱,流动相为乙腈0.1%磷酸,紫外检测波长为258nm。结果家兔灌服桃核承气汤和单味大黄后,大黄酸血浆浓度时间曲线符合二室开放模型。桃核承气汤中大黄酸吸收更为迅速,按相同给药剂量计(1mg/kg),灌服桃核承气汤和单味大黄大黄酸的Cmax分别为(0.682±0.142)mg/L、(0.358±0.105)mg/L,二者差异有显著意义(P<0.01)。结论桃核承气汤药味配伍促进了大黄酸的吸收利用。To study the pharmacokinetics of rhein after ig administration of Taohe Chengqi Decoction and rhubarb in rabbits. Methods: Normal rabbits were given ig a single dose of Taohe Chengqi Decoction 5g/kg or rhubarb 1.4g/kg. The plasma concentration of Rhein was measured by high performance liquid chromatography after extracted with perchloric acid and ether. Eclipse XDB-C18 was used as analytical column. The mobile phase consisted of acetonitrile-0.1% phosphoric acid, and the ultraviolet wavelength was set at 258nm. Results: The concentration-time curves of rhein fitted well into two-compartment models in rabbits. The results showed that rhein was absorbed more quickly after ig administration of Taohe Chengqi Decoction than rhubarb. With the same dosage of rhein(1 mg/kg) ig in rabbits, the maximum plasma concentration (Cmax) of rhein were (0.682±0.142) mg/L and (0.358±0.105) mg/L after ig administration of Taohe Chengqi Decoction and rhubarb. Significant difference was found between the two groups for the maximum plasma concentration (P<0.01). Conclusions: The compound compatibility of the decoction promoted the absorption of rhein. It indicated that there is rationality in the compatibility of Taohe Chengqi Decoction.
