摘 要:目的研究淫羊藿苷(icariin,ICA)伍用三七总皂苷(Panaxnotoginsengsaponins,PNS)对Aβ淀粉肽2535(betaamyloidpeptide2535,βAP2535)大鼠侧脑室注射空间学习和记忆障碍的影响。方法取经跳台法和八臂迷宫法筛选的正常大鼠采用侧脑室注射βAP2535,制成阿尔茨海默病(Alzheimer'sdisease,AD)大鼠模型,应用跳台法和八臂电迷宫法判断给药前后大鼠的空间学习和记忆能力;采用化学比色法测定脑组织中乙酰胆碱酯酶(acetylcholinesterase,AchE)活性。结果与溶剂组大鼠相比,模型大鼠八臂电迷宫错误次数明显增加(P<0.05),跳台学习和记忆错误次数明显增加(P<0.01)。大鼠造模前后连续igICA+PNS5+40,10+80mg/kg21d后,模型大鼠上述行为学指标得到明显改善(P<0.05),脑组织AchE活性降低(P<0.05)。结论ICA伍用PNS(5+40,10+80mg/kg)对βAP2535侧脑室注射所致大鼠空间学习和记忆障碍有显著的预防和治疗作用,该作用与降低脑组织中AchE活性呈相关性。OBJECTIVE To investigate the preventive and therapeutical effects of icariin (ICA) combined with the Panax notoginseng saponins (PNS) on spacial learning and memory dysfunction of Alzheimer's disease (AD) model in rats induced by β-amyloid peptide_ 25-35 (β-AP_ 25-35 ) lateral ventricle injection. METHODS After successfully trained by 8-arm radial electric maze and step-down passive avoidance test, the rats were administrated lateral ventricle injection with β-AP_ 25-35 followed by one week of drug pre- treatment. Spacial learning and memory ability were evaluated by 8-arm radial electric maze and step-down passive avoidance test after a period of drug therapy. The activity of acetylcholinesterase (AchE) in the brain was determined with visible spectrophotometry. RESULTS The learning and memory abilities of rats by lateral ventricle injection with β-AP_ 23-35 significantly weakened compared with those in the saline group through 8-arm radial electric maze test ( P <0.05) and step-down passive avoidance test ( P <0.01). The combination (ICA+PNS 5+40, 10+80mg/kg, ig) significantly protected and improved all the evaluated indexes of the model groups after three weeks ( P <0.05), moreover, reduced the activity of AchE in the brain ( P <0.05). CONCLUSION The combination can prevent and improve AD model rats established by lateral ventricle injection of β-AP_ 25-35 , which is related with inhibition of the AchE activity in the brain.
