摘 要:目的建立测定琥乙红霉素制剂含量的方法。方法利用琥乙红霉素与醌茜素在水乙醇介质中发生电荷转移反应,形成电荷转移络合物,采用可见分光光度法测定。结果荷移反应生成的荷移络合物在570nm处有最大吸收,表观摩尔吸光系数是1.14×104L·mol-1·cm-1,应用等摩尔连续变换法和斜率比法测得荷移络合物的组成比为1∶1,稳定常数是1.8×105。药物浓度在0~60mg·L-1范围内服从比耳定律,方法平均回收率在98.0%以上,RSD为0.82%(n=6)。结论方法稳定、准确、灵敏、快速,对样品的测定结果令人满意。OBJECTIVE To establish a method for rapid determination of erythromycin ethylsuccinate. METHODS To utilize visible spectrophotometry based on the charge transfer reaction between erythromycin ethylsuccinate as donor and quinalizarin as acceptor in water -alcohol solution. RESULTS The apparent molar absorptivity of complex was 1.14×10 4 L.mol -1 .cm -1 at 570 nm. The Composition ratio and stability constant of charge transfer complex was 1∶1 and 1.8 10 5 respectively by Job's and slope ratio method. Beer's law was obeyed in the range of 0~60mg·L -1 of erythromycin ethylsuccinate. The average recovery was over 98.0%. The average standard deviation of six determinations was 0.82 %. CONCLUSION The proposed method is stable, accurate, sensitive and rapid. It has been applied to the determination of erythromycin ethylsuccinate in tablets with satisfactory results.
