摘　　要：目的　探讨神经生长因子(NGF)在成年猴脊髓的表达分布情况,为成年猴脊髓存在此生长因子的表达提供免疫组织化学证据。方法　云南成年健康雄性猕猴5只,平均体重6kg左右,氯氨酮(0.25ml/kg)肌肉注射麻醉后,Zamboni液经心脏主动脉插管灌注固定动物,取T8 脊髓节段入 4%多聚甲醛后固定(4℃)6小时,在-20℃恒冷切片机下制作20μm厚的连续冰冻切片,间隔取片后分别以抗NGF抗体行免疫组化ABC染色。在Olympus光学显微镜下观察NGF在猴脊髓的表达分布情况。结果　1.NGF的免疫阳性反应产物在成年猴脊髓腹角、背角神经元均有分布。2. NGF的免疫阳性反应产物在猴脊髓背角 Lissauer束有分布,表达NGF的Lissauer束因染色而呈明显的毛刷状/发束状。3.NGF阳性神经膨体分布于猴脊髓背角Ⅰ、Ⅱ板层。4.NGF的免疫阳性反应产物在成年猴脊髓中央管(X板层)均有分布。结论　成年猴脊髓存在NGF的表达,提示其可能在成年猴脊髓的生理过程中发挥作用。Objective To explore the specific expression of Nerve Growth Factor(NGF)in the spinal cord of adult monkey. Methods Five healthy adult Rhesus macaques(male)of YunNan have been used in our experiment,whose mean weight is 6kg.After being anesthesied by muscle injection of Ketamine(0.25 ml/kg),these animals were sacrificed via a way of infusing Zamboni fluid into aorta with the help of aortic cannlation.Then fixed the T8 spinal cord taken out from the monkeys with 4%paraformaldehyd reinst for 6h(4℃).The spinal cord were made into frozen sections in 20μm thickness.Next,anti-NGF serum were applied with the immunohistochemical ABC method to detect the distribution of NGF in the spinal cord sections of adult monkey.At last,the distribution of NGF in the spinal cord was observed by the use of Olympus light microscope. Results 1.The immunoreactivity for NGF-like could be seen in both motoneurons and sensory neurons in spinal cord of the adult monkey.2.The NGF-like immunoreactivities could,also be seen in spinal of the adult monkey.3.The distribution of NGF could be detected in the Lissauer tract of the adult monkey,as a result,the positive staining of Lissauer tract looked like hairbrush.4.The positive distribution of NGF could be seen in the,spinal lamina Ⅰ/Ⅱ of the adult monkey.5.The NGF-like immunoreactivity could be found in the spinal central canal of the adult monkey. Conclusions We provide the morphological evidences that there exists the expression of NGF in the spinal cord of adult monkey.This indicated that NGF may be play multifunctional and complex roles in the spinal cord of adult monkey.
