慢性重型肝炎患者血浆对C3A 细胞增殖功能的影响

摘　　要：目的观察慢性重型肝炎患者血浆对 C3A 细胞株增殖功能的影响,为研究应用 C3A 细胞株的生物反应器进行生物人工肝支持治疗提供实验依据。方法分别用100%正常人血浆(normal human plasma,NHP)和100%慢性重型肝炎患者血浆(chronic severe hepatitis plasma,CSHP)培养 C3A 细胞,通过四甲基偶氮唑盐法(methyl thiazolyl tetrazolium,MTr)检测细胞活力,光镜下观察 C3A 细胞在100% CSHP 中和在100% NHP 中的状态,扫描电镜观察细胞在100% CSHP 中和在100% NHP 中的生长状态,研究慢性重型肝炎患者血浆对 C3A 细胞增殖的影响。结果①连续5d 检测100% CSHP 培养的 C3A 细胞活力均明显高于100% NHP 组(t 值分别为6.26、4.89、3.55、3.94、6.17,P 值均小于0.01);②姬姆萨染色可见100% CSHP 培养的C3A 细胞生长较密;③48h 电镜下可见2组细胞生长状态均良好,但是100% CSHP 组处于分裂前期细胞数明显多于100%NHP 组,且分裂较活跃。结论 CSHP 对C3A 细胞的增殖有促进作用。Objective To investigate the effects of plasma from patients with chronic severe hepatitis on C3A cell line in vitro to which can provide experimental data for C3A cell derived bioartificial liver.Methods C3A cells were cultured in normal human plasma (NHP)and chronic severe hepatitis plasma(CSHP).We detected the cell activity by MTT and cellular morphosis by inverted micro- scope and scanning electron microscope to elucidate the effect of chronic severe hepatitis plasma on C3A ceils.Results ①The activities of C3A cells cultured in CSHP were higher than those in NHP for successive 5 days(P<0.01 ).②With inverted microscope,the C3A cells cultured in CHSP for 24 hours looked thicker than those in NHP.③The observation with scanning electron microscope demonstra- ted that the C3A cells grew well after being cultured for 48 hours in both groups,while the cells in prophase of division in CHSP were much more than those in NHP.Conclusion CHSP can promote the proliferation of C3A cell.
