CpGODN联合乙肝疫苗对小鼠免疫应答效果的影响

摘　　要：目的观察CpGODN联合乙肝疫苗免疫小鼠后,对小鼠的特异性免疫应答效果的影响。方法不同剂量乙肝疫苗单用或联合CpGODN免疫小鼠,用ELISA方法检测抗HBs水平,流式细胞仪检测外周血CD4+和CD8+T淋巴细胞亚群。结果免疫2周后,CpGODN联合中、高剂量乙肝疫苗组抗HBs水平较单用乙肝疫苗组高(P<0.05);免疫4周后CpGODN联合中、高剂量乙肝疫苗组抗HBs水平显著高于联合低剂量乙肝疫苗组(P<0.05),CpGODN联合低、中剂量乙肝疫苗抗HBs水平较单用乙肝疫苗明显升高(P<0.05),且以CpGODN联合中等量乙肝疫苗产生的抗HBs水平最高;T淋巴细胞亚群分析显示,联合组CD4+T淋巴细胞百分比明显高于单用乙肝疫苗组(P<0.05),CD4+/CD8+比值联合组也明显高于单用乙肝疫苗组(P<0.05)。结论CpGODN联合乙肝疫苗不仅能诱导出高水平的抗HBs,而且对细胞免疫也有调节作用。Objective To investigate the effects of CpG ODN on immune respones in mice with hepatitis B vaccine. Methods Different doses of hepatitis B vaccine with or without CpG ODN were injected to immune mice. Serum hepatitis B surface antibody(anti-HBs) levels in mice were detected by ELISA assays in two weeks and four weeks after injection. Changes of peripheral blood T lymphocyte subsets CD_4^+、 CD_8^+ and CD_4^+/CD_8^+ ratio were detected by flow cytometry. Results ①After two weeks by injection, Serum anti-HBs levels in mice injected by CpG ODN combining with hepatitis B vaccine were higher than that of the mice only injected by hepatitis B vaccine, P<0.05. ②After four weeks, Serum anti-HBs levels in mice injected with CpG ODN combining with 0.75 μg and 1.0 μg doses of hepatitis B vaccine were much higher than that combining with 0.25 μg doses of hepatitis B vaccine. ③After four weeks, compared with that of the mice only injected by hepatitis B vaccine, serum anti-HBs levels in mice with CpG ODN combining with 0.25 μg and 0.75 μg hepatitis B vaccine were significant higher except in mice with CpG ODN combining with 1.0 μg hepatitis b vaccine. ④Percentages of CD_4^+ of mice immunized with CpG ODN combining with hepatitis B vaccine increased more than that only with hepatitis b vaccine. Conclusion CpG ODN combining with hepatitis B vaccine not only induces the high level of antiHBs,but also can enhance the cellularimmune response.
