兔眼局部应用氧氟沙星和妥布霉素的房水移行性实验研究

摘　　要：目的比较氧氟沙星和妥布霉素在兔眼局部使用的房水移行性。方法取28只家兔随机分成2组。分别于滴眼后10、20、30、40、60、90、120分钟后抽取房水。右眼点氧氟沙星,左眼点妥布霉素。另取7只兔作为空白对照组,给予生理盐水滴眼。采用高效液相法检测房水内的药物浓度。结果滴眼后兔前房内的药物浓度高峰值出现时间,氧氟沙星在滴眼后30～40min,妥布霉素在滴眼后20min左右。参照各自的MIC90值,氧氟沙星在给药后20min时房水浓度达到MIC90,可持续到60min。结论氧氟沙星具有良好的房水移行性,房水内药物浓度高,持续时间长。Objective To compare the anterior aqueous humor levels of ofloxacin and tobramycin following topical ocular administered in rabbits.Methods 28 healthy rabbits were randomly divided into two groups in different application modes. Rabbits were dosed with single drop and aqueous humor were collected in different time. Ofloxacin were administered in right eyes and tobramycin in left eyes.7 rabbits were used as control group and dosed with saline. High performance liquid chromatography was used to analyze the drug level.Results The peak concentrations of ofloxacin and tobramycin is at 30～40 and 20 minutes after administered respectively. The concentrations of ofloxacin are higher than the MIC_ 90 even 60 minute after last dosing in single dose mode, but the concentration of tobrymycin in aqueous humor are all lower than its MIC_ 90.Conclusion Ofloxacin concentrations in aqueous humor reached high levels and remained long period in healthy rabbits. Ofloxacin may be useful agent in preventing and treating endophthalmitis.
