摘  要:触珠蛋白(Haptoglobin简称Hp)是一种存在于人类及其他哺乳动物血清和其他体液中具有遗传多态性的X2糖蛋白。S—mithies首先发现了Hp的遗传多态性,用淀粉凝胶电际技术将其分为三种表现型。嗣后,Smithies与Connell又将Hp分为六种表现型。进入八十年代,Shibata与Mastumoto等人采用等电聚焦电泳技术将Hp进一步分为16种表现型,We examined the Hp subtypes both in the human sera and in the dried bloodstains using a method for Hp purifacation, the hydrolysis of Hp performed using the β-merca- ptoethanol-ureaiodacetamid mixture and the isoelectric focusing electrophoresis on slab polyacrylamide gel. Hp phenotyping of 404 serum samples from unrelated chinese blood donors in Xian were carried out. The distribution of the Hp subtypes are: Hp^(2SS)0.25%, Hp^2F^S49.5%, Hp^(1S)6.68%, Hp^(2fS-2SS)4.21%, Hp^(2SS)-^(1S)1.49%, Hp^(2fs)-^(1S)35.89%, Hp^(2fS)-^(1S)0.495%, Hp^2-~S-^(2V)0.743%, Hp^(1S)-^(1f)0.25% and Hp^(2V)-^(1S)0.495%. The gene frequencies respectively are: Hp^(1f)0.0037, Hp^(1S)0.2574, Hp^(2fS)0.0309, Hp^(2fS)0.7017 and Hp^(2V)0.0063. The observed and expected values provided a good fit to the Hardy-Weinberg equilibrium(p>0.05). The probab ility of excluding paternity means 0.2601, and discrimination probability means 0.62. The 24 samples of bloodstains were kept in room tempreture for seven days, 4℃ for seven days and -30℃ for thirty days. In addition to three samples, the others could besubtyped agreeing with the subtype of Hp in their serum.
