摘  要:目的研究含去痴灵水提物血清对M146L细胞分泌的β淀粉样蛋白(βamyloidprotein)的影响,探讨去痴灵治疗阿尔茨海默病(Alzheimer’sdisease,AD)的可能性及作用机制。方法采用血清药理学方法,制备含去痴灵水提物血清。体外培养稳定转染人类阿尔茨海默病β淀粉样前体蛋白(betaamyloidproteinprecursor,APP)和突变型早老素1(prsenilin1,PS1)基因的中华仓鼠卵巢上皮细胞系(M146L),使之高效产生β淀粉样蛋白42(Aβ42),建立Aβ42过度表达的细胞模型。加入含去痴灵水提物血清,用MTT法检测不同浓度(12%、6%、3%)的含药血清对M146L细胞毒作用,用ELISA法测定其对M146L细胞分泌Aβ42的影响。结果3个浓度的含药血清对M146L存活均无影响,对细胞分泌的Aβ42有抑制作用,并呈现一定的量效关系。结论一定剂量(12%、6%、3%)的含去痴灵水提物血清对M146L细胞分泌的Aβ42有明显的抑制作用,其机制尚待进一步研究。Objective: To approach the possibility and mechanism that Quchiling treats Alzheimer’s disease(AD), we studied the effect of Quchiling water extract-contained serum(QCS) on the secretion of β-amyloid protein 42(Aβ_ 42 )by Chinese hamster ovary cells which stably express β-amyloid protein precursor(APP) and mutant preseinlin 1(M146L). Methods: The QCS was prepared using rats. The cell model, M146L, producing β-amyloid protein in vitro was established by transfecting APP and mutant presenilin-1 genes. The mononuclear cell directed cytotoxicity assay(MTT) test and enzyme linked immunosorbent assay(ELISA) were performed to evaluate the effect of the QCS on the cell model and to analyse Aβ_ 42 secretion. Results: The MTT test indicated that different concentrations of the QCS had no effect on the cultured M146L cell survival. And the ELISA test showed that the secretion of Aβ_ 42 by the M146L cell decreased obviously. Conclusion: The suitable dose of the QCS can inhibit the secretion of Aβ_ 42 by M146L cell, and its mechanism will be further studied.
