甲基莲心碱增强环磷酰胺诱导小鼠Lewis肺癌细胞凋亡的作用

摘　　要：目的评价甲基莲心碱(Nef)对环磷酰胺(CTX)诱导小鼠Lewis肺癌细胞凋亡作用的影响。方法Lewis肺癌移植瘤小鼠40只,雄性,体重18～22g,随机分成CTX+Nef、CTX、Nef、NS4个组,每组10只。通过检测瘤重抑制率及瘤细胞凋亡率,观察Nef联合CTX对小鼠Lewis肺癌的疗效和诱导癌细胞凋亡的影响。结果CTX+Nef、CTX组的瘤重抑制率分别为80.08%、58.95%,明显高于Nef组(6.78%),P<0.01;而CTX+Nef组较CTX组也有增高(P<0.05);流式细胞术检测CTX+Nef、CTX组的癌细胞凋亡率分别为53.50%、25.92%,明显高于Nef、NS组(7.09%,5.63%),P<0.01;CTX+Nef组也高于CTX组(P<0.05);TUNEL法检测CTX+Nef、CTX组的癌细胞凋亡指数分别为(10.24%±11.08%),(5.36%±6.24%),较Nef、NS组明显增加(1.45%±1.98%,1.03%±1.65%),P<0.05;CTX+Nef组的凋亡指数较CTX组也有明显增加(P<0.05)。结论甲基莲心碱能增强环磷酰胺诱导的小鼠Lewis肺癌细胞凋亡作用,且两者具有协同效应。Objective To investigate the effect of Neferina on cell apoptosis induced by cyclophosphamide in mice Lewis Lung Carcinoma.Methods 40 male mice with inoculated Lewis Lung Carcinoma were divided randomly into 4 groups to receive either CTX+Nef,or CTX,or Nef,or nothing.The growth inhibitory rate and cancer cell apoptosis were detected by PI stain flow cytometry and TUNEL assay respectively.Results The difference in growth inhibitory rate were significant between CTX+Nef group(80.08%) and CTX group(58.95%)(P<0.05),or between CTX+Nef group(80.08%) and Nef group(6.78%)(P<0.01),or between CTX group(58.95%) and Nef group(6.78%)(P<0.01).Flow cytometry showed that cell apoptosis rate in CTX+Nef(53.50%)、CTX group(25.92%) were markedly higher than that in Nef and NS group(7.09%、5.63%)(P<0.01).The cell apoptosis was significantly different between CTX+Nef group and CTX group(P<0.05).The exponent of cell apoptosis detected by TUNEL assay were apparently increased in CTX+Nef(10.24%±11.08%),CTX group(5.36%±6.24%),as compared with that in Nef and NS group(1.45%±1.98%、1.03%±1.65%,P<0.05).The exponent of cell apoptosis was also markedly higher in CTX+Nef group than in CTX group(P<0.05).Conclusion Nef has a synergetic effect on cell apoptosis induced by cyclophosphamide in mice Lewis Lung Carcinoma.
