荧光染料CFSE检测淋巴细胞增殖的研究

摘　　要：目的　评价荧光染料羟基荧光素二醋酸盐琥珀酰亚胺脂(CFSE)用于淋巴细胞及其亚群增殖状况追踪的效果。方法　利用荧光染料CFSE作为细胞标记,结合荧光标记的抗体,通过流式细胞仪检测淋巴细胞及其亚群体外增殖情况。结果该方法不仅可以测定淋巴细胞及其亚群的增殖情况,而且可以估算细胞分裂的周期。结论　荧光染料CFSE是一种可以监测淋巴细胞及其亚群增殖状况的很好的示踪剂。Objective To evaluate the role of carboxyfluorescein diacetate succinimidyl ester (CFSE) in the determination of lymphocyte proliferation.Methods CFSE staining combined with fluorescent antibody labeling and flow cytometry are used to detect the proliferation kinetics of lymphocytes and their subsets after stimulation of colonel stimulators in vitro.Results The results show that this technique is not only applicable to proliferation analysis of lymphocytes and their subsets,but can estimate the rounds of cell division using flow cytometry.Conclusion CFSE is an excellent stain to be used to analysis the lymphocytes and their subsets proliferation.
