摘  要:目的 探讨NM - 3及氧化苦参碱对裸鼠体内多药耐药人胃癌细胞SGC - 790 1的抑制作用及其抗肿瘤作用的分子机制。方法 建立多药耐药人胃癌SGC - 790 1细胞的皮下移植瘤模型并随机分成生理盐水对照组,NM - 3、氧化苦参碱、NM - 3联合氧化苦参碱治疗组。每组裸鼠在胃癌组织移植7周后处死,并测算其肿瘤抑制率和采用TUNEL(末端脱氧核酸转移酶原位标记)和流式细胞仪法分析其凋亡指数。结果 NM - 3治疗组( 10mg/L、2 0mg/L)对多药耐药SGC - 790 1细胞皮下移植瘤的抑制率显著增加,其凋亡率分别是( 11.2 0±0 .18) %和( 12 .2 0±1.6 5 ) % ,两者比较无差异( P >0 .0 5 ) ;NM - 3联合氧化苦参碱治疗组凋亡指数(AI)为( 5 1.2 0±4 .2 3) % ,显著高于NM - 3治疗组和生理盐水对照组( 1.2 7±0 .11% ,P <0 .0 1)。结论 NM - 3可以增强氧化苦参碱对多药耐药细胞SGC - 790 1的抑制作用,诱导裸鼠体内人胃癌细胞的凋亡。Objective To study molecular mechanisms of the effects of NM-3 and oxymatrine on cultured multidrug-resistant gastric cancer SGC-7901 cells in nude mice with xenografted human gastric cancer.Methods Mouse model of subcutaneously transplanted multidrug-resistant human gastric cancer SGC-7901 cells was developed.The model nude mice were divided randomly into four main groups: normal saline control group,NM-3 group,oxymatrine group,and NM-3 plus oxymatrine group.Seven weeks after implantation,the mice were sacrificed,their tumor inhibiting rates were determined, and the apoptotic indices of multidrug-resistant SGC-7901 cells in vitro and the apoptotic indices of gastric cancer cells were assayed by using TUNEL method and flow cytometry.Results The tumor inhibiting rate in NM-3 group was significantly increased,and the apoptotic rates in the 10 mg/L and 20 mg/L dosage subgroups were (11.20±0.18)% and (12.20± 1.65 )%,respectively,without significant difference between the two subgroups. The AI in NM-3 plus oxymatrine group was (51.20± 4.23 )%,significantly higher than those of NM-3 group and saline control group(1.27±0.11%,P<0.01).Conclusions NM-3 enhances the inhibitory effects of oxymatrine on multidrug-resistant SGC-7901 cells and induces apoptosis of human gastric cancer cells in nude mice. The results suggest that NM-3 enhances the inhibitory effects and sensitivity of chemotherapy for human gastric cancer in vivo.
