摘 要:目的 建立骨髓基质干细胞(marrowstromalcell,MSC)内耳移植技术,研究内耳细胞移植的可行性。方法 将豚鼠骨髓基质干细胞分离培养,用4 ,6 -联脒- 2 -苯基吲哚( 4 ,6 -diamidino - 2 -phenylindole,DAPI)荧光标记,采用显微注射技术在耳蜗底周鼓阶钻孔,通过钻孔处缓缓注入细胞,术后14天处死,行耳蜗石腊切片,荧光显微镜及HE染色观察移植细胞成活情况。结果 骨髓基质干细胞内耳移植成活,并贴壁生长。结论 成功地建立了骨髓基质干细胞内耳移植技术。Objective To establish the method of marrow stromal cell(MSC) transplantation into the guinea pig cochlea and analyze the feasibility of MSC transplantation.Methods MSCs were isolated from marrow of the guinea pigs and cultured in vitro. MSCs labeled with DAPI were transplanted into the guinea pig cochlea via a small hole drilled on the bony wall of tympanic scala in the basal turn of the cochlea.10 μl cell suspension was inoculated into the perilymphatic space, and artificial perilymphatic fluid (APF) was injected as a negative control.After 14 days the animals were sacrificed and the otic bulla was harvested, paraffin-embedded, and cut in a paramodiolar plane. The transplant-derived cells were examined with fluorescent microscope. The histopathological changes were observed by HE staining.Results All transplant-derived cells survived in the perilymphatic space of the cochlea, and gradually attached to the bone wall.Conclusion MSC can survive in the inner ear after transplantation,which provide a basis for study of MSC differentiation and expression in the inner ear.
