摘  要:目的观察吡咯二硫氨基甲酸酯(PDTC)对体外循环中心肌的保护效果。方法12只犬随机分为对照组(C组)和PDTC组(P组),每组6只。P组于CPB转机前静脉注入PDTC30mg/kg,C组静脉注入等容量生理盐水。两组均全心缺血60min,恢复灌注60min。于并循5min、阻断60min及开放60min观察心机复跳情况,并测定心脏指数(CI)的变化;于开放60min取心肌观察形态学结构。结果①开放升主动脉后,C组1例自动复跳,而P组3例自动复跳;②C组与P组在开放30min后,CI均恢复缓慢,但组间比较P组恢复较C组快(P<0.05);③光镜下心肌形态学观察,P组在开放60min时心肌结构保存良好。结论PDTC对体外循环中缺血心肌具有良好的保护效果。Objective To evaluate the myocardial protective effect of pyrrolidine dithiocarbamate, an agent that inhibits the nuclear localization of the transcription factor nuclear factor (NF-κB),on ischemic myocardium during cardiopulmonary bypass.Methods Twelve adult hybrid health dogs were randomly divided into PDTC group (group P, n=6) and control group(group C,n=6).After tracheal intubation,PDTC 30mg/kg was administered intravenously in group P,while in group C only equivalent of normal saline was administration.The heart was subjected to 60 min global ischemia followed by 30 min reperfusion. Hemodynamics (CI) was measured before aortic cross-clamping and 30min,60min after aortic unclamping. Myocardial tissue was taken at 60min after reperfusion for structure examination with optional microscope.Results Hemodynamics recovery after aortic declamping was significantly better in group P than that in group C. Myocardial structure was less injuried in group P.Conclusions Pretreatment with PDTC is effective in reducin myocardial injury during cardiopulmonary bypass.
