摘  要:选用体重55kg左右的肥育后期杜×(长×梅)三元商品猪80头,随机分成实验组和对照组,实验组在基础日粮中添加600mg/kg半胱胺盐酸盐。35d后进行屠宰实验,并采集血样、背部皮下脂肪组织和下丘脑。用放射免疫法测定血清胰岛素(insulin)、瘦素(leptin)和肿瘤坏死因子α(TNF-α)浓度,用RT-PCR方法,以18SrRNA作内标,定量分析脂肪组织中生长激素受体(GH-R)、胰岛素样生长因子-1(IGF-1)、胰岛素样生长因子I型受体(IGF-IR)和leptinmRNA丰度以及下丘脑Ob基因b型受体(Ob-Rb)基因表达。结果显示:半胱胺分别使猪的体重和日增重提高1.54%(P<0.05)和4.83%(P<0.05),分别使胴体脂率和6、7肋背膘厚下降14.90%(P<0.05)和20.14%(P<0.05)。实验组血清中Leptin水平下降39.33%(P<0.05),血清中TNF-α水平升高25.00%(P<0.05),但血清中insulin水平在二组间没有差异。实验猪脂肪细胞leptinmRNA丰度比对照猪减少15.02%(P<0.05);实验猪脂肪细胞GH-R、IGF-1、IGF-IRmRNA丰度以及下丘脑leptin-RbmRNA丰度与对照猪无显著差异。表明半胱胺可抑制猪肥育后期的体脂沉积,改善胴体品质,而这种作用并不是通过改变脂肪组织中GH-R、IGF-1和IGF-IR的表达来实现的。Eighty Duroc-Jersey × (Landrace×Meishan) finishing pigs weighting 55 kg were allotted into treatment and control groups. The treatment group was fed with the diet supplemented with 600 mg/kg cysteamine for 35 d . Pigs were slaughtered for the blood, dorsal subcutaneous adipose tissue and hypothalamus sampling. The serum concentrations of insulin, leptin and TNF-α were determined by RIA, the total RNA was extracted from tissues to measure the abundance of the growth hormone receptor (GH-R), insulin-like growth factor 1 (IGF-1), insulin-like growth factor I receptor (IGF-IR), Leptin mRNA in adipose tissue and Ob-Rb mRNA in hypothalamus by RT-PCR using 18S rRNA as an internal standard. Results showed that the cysteamine enhanced the body weight and average daily weight gained by 1.54% (P < 0.05) and 4.82% (P < 0.05), decreased the adiposity index and backfat thickness in 6 and 7th rib by 14.90% (P < 0.05) and 20.14% (P < 0.05), decreased the serum leptin concentration by 39.33% (P <0.05), increased serum TNF-α by 25.00% (P < 0.05). Cysteamine down-regulated the relative abundance of leptin mRNA in dorsal subcutaneous adipose tissue by 15.02% (P < 0.05). However, no differences in serum insulin level, the relative abundance of GH-R, IGF-1 and IGF-IR mRNA in dorsal subcutaneous adipose tissue and Ob-Rb mRNA in hypothalamus between cysteamine treated and control group were found. These results suggested that cysteamine inhibitted backfat deposition and improved carcass quality in the finishing pigs, while not altering the expression of GH-R, IGF-1 and IGF-IR in the adipose tissue.
