农杆菌介导的沿海甜菜(Beta maritima L.)遗传转化条件的优化

摘  要:采用农杆菌介导法,将外源基因导入离体培养的沿海甜菜丛生芽芽尖中并再生植株。分析了芽尖大小、生理状态、浸染液浓度、浸染时间、共培养天数、负压处理和乙酰丁香酮的浓度对抗性芽尖比率的影响。在农杆菌浸染时,用负压处理(40kPa)6min、添加100μM乙酰丁香酮得到高达34%的抗性芽尖比率。The shoot tips from shoot clumps of sea beet (Beta maritima L.) were transformed with Agrobacterium tumefaciens,and fertile plantlets were regenerated from the transformed shoots. The shoot tips size,the concentration of Agrobacterium suspension,period of immersing time,period of co-culture,vacuum infiltration,and the concentration of acetosyingone (AS) were respectively evaluated during the process of infection. The highest transformation efficiency was obtained to 34% when 2-3 mm shoot tips were infected in Agrobacterium suspension (supplemented 100μM AS) of OD600 0.3,vacuum infiltrated at 40 kPa for 6 min,and then co-cultured for 3-4 d.
