摘 要:目的:探讨瘦素(leptin)、纤连蛋白(FN)对细胞滋养细胞浸润能力的影响及其协同作用。方法:①取孕6-8周的人工流产妊娠绒毛,分离、培养细胞滋养细胞并经光镜、电镜、免疫细胞化学鉴定。②用RT-PCR方法检测细胞滋养细胞中瘦素的表达。③应用体外培养侵袭模型检测瘦素、FN及二者协同时对细胞滋养细胞浸润能力的影响。结果:①分离、纯化所得细胞滋养细胞有瘦素mRNA表达。②瘦素组、FN组和瘦素与FN协同组浸润穿过matrigel的滋养细胞数均显著多于对照组(P<0.01),其中瘦素与FN协同组滋养细胞浸润能力最强。结论:①瘦素能增强细胞滋养细胞的侵袭能力。②FN对细胞滋养细胞的浸润具有趋化作用。③瘦素与FN对细胞滋养细胞的浸润有正协同作用。Objective: To study the effects of leptin, fibronectin and their synergy on cytotrophoblast invasion activity. Methods: Cytotrophoblasts (CTB) were isolated from chorionic villi at 6-8 weeks pregnancy, evaluated by light, electron microscope and immunocytochemistry and cultured in vitro. Using transwell invasion model coated with matrigel, the change of cytotrophoblast invasion ability was observed before/after adding leptin, FN or leptin and FN into culture medium. Results: The isolated cells cytotrophoblasts and they expressed leptin mRNA. The number of penetrated-membrane-cells in leptin, FN, leptin + FN groups were significantly higher than control group. The number of invasive CTB in leptin+FN group was larger than that in leptin group or FN group. Conclusions: Leptin can increase the invasion ability of cytotrophoblast. FN can promote the invasion ability of trophoblasts. Leptin-FN synergy can significantly enhance the CTB invasion ability than FN or leptin alone. It proves that CTB invasion procedure is modulated by multiple factors of cell-to-cell and cell-to-matrix interaction.
