摘　　要：目的研究红细胞趋化因子受体(ECKR)在寻常型银屑病患者外周血中性粒细胞中的表达情况,探讨其在银屑病发病机制中的作用。方法应用流式细胞仪和特异性FITC标记的鼠抗人ECKR抗体检测寻常型银屑病患者和正常人外周血中性粒细胞表面ECKR蛋白的表达水平。应用逆转录-聚合酶链反应(RT-PCR)方法检测28例寻常型银屑病患者以及30例正常人外周血中中性粒细胞内ECKRmRNA的表达。并将结果与患者皮损面积及严重程度指数(PA-SI)进行了相关性分析。结果寻常型银屑病患者外周血中性粒细胞膜ECKR阳性率明显低于正常人,进行期患者较稳定期患者显著降低,且患者中性粒细胞膜ECKR的阳性率与其PASI评分呈负相关。患者外周血中性粒细胞内ECKRmR-NA的表达水平明显高于正常人,而进行期患者ECKRmRNA的表达又高于稳定期患者。结论ECKR在银屑病发病过程中发挥一定的作用,其表达水平与疾病的进展密切相关。Objective To determine the expression of erythrocyte chemokine receptor (ECKR) on peripheral blood neutrophils of patients with psoriasis vulgaris and to explore its clinical significance.Methods ECKR on peripheral blood neutrophils was detected by direct immunofluorescence staining method. ECKR on neutrophils,which combined with FITC-labeled ECKR mAb,was detected by flow cytometry. Reverse transcription-polymerase chain reaction (RT-PCR) technique was applied to semi-quantatatively analyze ECKR mRNA expression in peripheral blood neutrophils from 28 cases of psoriatic patients,and 30 healthy controls.The correlation between ECKR and psoriatic aera and severity index (PASI) was evaluated.Results The results showed that the positive rate of and ECKR+ neutrophils in psoriasis was significantly lower than that in the controls (P<0.01).The positive rate of ECKR+ neutrophils in active psoriasis was lower than that of resting psoriasis.Furthermore,the positive rate of ECKR+ neutrophils in psoriasis was closely related with PASI score of psoriasis.ECKR mRNA levels in neutrophils from psoriatic patients were markedly higher than those in normal controls.Conclusion ECKR may be regarded as clinical reference index to observe the change of the disease progress.
