摘 要:目的: 观察慢性膀胱出口梗阻后P2X1受体的表达水平.方法: 雌性Wistar大鼠70只,体质量250~300 g,40只为实验组,30只同龄雌性Wistar大鼠为对照组.在部分膀胱出口梗阻的动物模型成功建立后8 wk开始试验,采用免疫组织化学方法结合IMAGE-PRO图像处理软件对大鼠膀胱P2X1的表达(即阳性颗粒在某个视野下所占的百分比)进行定量研究.结果: 部分膀胱出口梗阻后大鼠较对照组大鼠膀胱的体积增大,黏膜层增厚,P2X1受体的表达水平平均为在膀胱平滑肌层[(9.60±4.78) vs (2.31±1.19)] (P<0.01),在膀胱黏膜层[(0.12±0.03) vs (0.06±0.02)] (P<0.01),浆膜层[(0.04±0.02) vs (0.03±0.01)] (P>0.05),尿道[(6.05±2.25) vs (1.48±0.14)] (P<0.01),慢性膀胱出口梗阻后P2X1受体的表达水平均明显高于对照组.结论: 膀胱出口部分梗阻后,P2X1受体在膀胱各个部位表达均升高.推测P2X1可能参与了慢性膀胱出口梗阻后膀胱的生理或病理功能的调节.AIM: To study the expression of purinoceptor P2X_1 subunit after partial bladder outlet obstruction in rats.METHODS: Partial bladder outlet obstruction (BOO) was induced in 250~300 g female Wistar rats (n_1=40 in experimental group).Sham operated age-matched rats were taken as controls (n_2=30 in control group).Eight weeks post operation,the expression of P2X_1 in bladder was determined with immunohistochemisty and IMAGE-PRO image editing software and was compared with that of control group.RESULTS: Eight weeks after BOO,the volume of bladder increased and the mucosa was thicker compared with that in the control group.The level of purinoceptor P2X_1 expression in BOO group was significantly higher than that in control group [BOO group vs control group: (9.60±4.78) vs (2.31±1.19) in bladder smooth muscle (P<0.01),(0.12±0.03) vs (0.06±0.02) in the bladder mucosa (P>0.05),(0.04±0.02) vs (0.03±0.01) in the seroma (P>0.05) and (6.05±2.25) vs (1.48±0.14) in the urethra (P<0.01)].CONCLUSION: After the bladder outlet obstruction,the level of purinoceptor P2X_1 increases in the bladder,suggesting that the up-regulation of P2X_1 in bladder is involved in the regulation of physiological and pathophysiological function following the chronic bladder outlet obstruction in rats.
