摘 要:目的:研究Ⅰ、Ⅲ型前胶原核酶治疗裸鼠增生性瘢痕后胶原含量及分布的改变。方法:①建立裸鼠增生性瘢痕动物模型(80只)。②将裸鼠随机分为:实验组(A组20只,注射Ⅰ型前胶原核酶;B组20只,注射Ⅲ型前胶原核酶;C组20只,注射Ⅰ型及Ⅲ型前胶原核酶;各组又分为2. 5μg/100μL及5. 0μg/μL两个剂量组,每个剂量组各10只裸鼠,每天用微量注射器行瘢痕内多位点注射1次,共7d)及对照组(D组10只,注射生理盐水7d;E组10只,空白对照组)。③应用苦味酸天狼猩红染色偏振光法,结合图像分析技术,观察分析各组胶原含量及分布的改变。结果:A组Ⅰ型胶原由紧密排列,变成较稀疏,提示含量减少;B组Ⅲ型胶原在血管旁及Ⅰ型胶原间分布减少;C组Ⅰ、Ⅲ型胶原均减少;D组和E组无明显变化。结论:Ⅰ、Ⅲ型前胶原核酶治疗裸鼠增生性瘢痕后,可使胶原的排列分布稀疏,含量减少,形态改变明显。Objective:To study the morphological changes of collagen in human hypertrophic scar in a nude mice model treated with ribozymes. Methods:To establish a model of human hypertrophic scar in nude mice(80 mice).All the mice were randomly divided into four groups:group A(typeⅠprocollagen ribozyme treated,20 mice), group B(type Ⅲ procollagen ribozyme treated,20 mice),group C(typeⅠand Ⅲ procollagen ribozyme treated,20 mice),group D(normal saline solution injected,10 mice)and group E(blank control,10 mice). Tow subgroups were established by microinjection of 2.5 and 5.0μg/100 μL of ribozymes of type Ⅰ and Ⅲ procollagen in groups A,B and C for 7 days. The sirius red staining and polarization microscopy, combined with image analysis were applied to observe the results. Results:The ribozymes can inhibit type Ⅰand Ⅲ collagen of hypertrophic scar. Under the polarization microscopy, the structure of scar became loose, the content and distribution of collagenous fibre diminished. There were no remarkable changes in group D and group E. Conclusions:The results suggested that ribozymes of type Ⅰand Ⅲ procollagen can inhibit collagen production, which was proved morphologically.
