摘 要:目的对我国登革2型病毒1998年广东省南海市流行株GD08/98结构蛋白基因进行序列测定及分析,为追踪其地域来源、基因组结构与致病性的关系及研究开发登革病毒新型疫苗奠定基础。方法根据登革2型国际标准株NGC序列,设计2对重叠引物,RT—PCR扩增,分别克隆到pMD18—T载体,转化受体菌DH5α,挑取阳性克隆进行PCR、酶切鉴定及序列测定。结果此登革病毒株结构蛋白基因序列长度为2325bp,编码775个氨基酸。与其它登革2型病毒株ThNH81/93、NGC、44、TSV01、04及S1进行比较,其核酸序列同源性分别为98%、94%、94%、92%、92%、90%。结论GD08/98病毒株的结构基因序列基本类似于已发表的其它登革2型病毒株。GD08/98株与ThNH81/93株同源性最高,推测其可能来源于泰国。Objective Through sequencing and structural analysis of the structural protein genes of dengue virus type 2 strain GD08/98 isolated from Nanhai, Guangdong province, providing bases for the molecular epidemiology study of dengue virus and potential vaccine selection for Chinese dengue virus isolates. Methods Consulted dengue virus type 2 NGC strain, two pairs of primers were designed. The structural protein gene of dengue virus type 2 strain GD08/98 was amplified by reverse transcriptase-polymerase chain reaction (RT PCR), then cloned into pMD18-T vector and sequenced. Results The structural protein genes of strain GD08/98 constitutes by 2325 nucleotides and encoding 775 amino acids. The nucleotide homology compared with strain ThNH-81/93(Thailand 1993) was 98%, compared with strain 44(China) and NGC (New Guinea-C) were both 94%, compared with strain 04(China) and TSV01(Australia 1993) were both 92%, compared with strain Sl was 90%. Conclusions The sequence of dengue virus type 2 strain GD08/98 structural protein genes are identical to the published dengue virus type 2 sequence. GD08/98 show a highest homology with ThNH81/93. Phylogenetic analysis suggests that GD01/98 might come from Thailand.
