摘  要:目的:建立分光光度法直接测定壳聚糖的含量。方法:利用壳聚糖分子中的伯氨基与茚三酮在酸性条件下生成有色缩合产物,进行比色测定,并对影响比色反应的因素进行分析,优化分析条件。结果:缩合产物在570nm 有最大吸收,优化条件下制备的壳聚糖标准曲线方程为:A=0.022C-0.0196,r=0.9999,线性范围为4.00-40.00μg·mL^(-1),平均回收率为99.96%,RSD=1.1%(n=9)。结论:茚三酮比色法测定壳聚糖含量,方法简便快捷,重现性好,灵敏度高,可以用做壳聚糖的定量方法。Objective:A spectrophotometric method for quantitative analysis of chitosan based on the ninhydrin reac- tion were proposed.The effects on this reaction were also investigated.Methods:The reaction of chitosan with nin- hydrin under a specified condition will form a colored reaction product,which can be measured at 570 nm on UV- VIS spectrophotometer.Results:The dominent factors for the ninhydrin reaction include molecular mass of chitosan, heating time,the pH and concentration of acetate buffer,the concentration of ninhydrin reagent.The calibration curve equation was linear in the range 4.00-40.00μg·mL^(-1) of chitosan,A=0.022C-0.0196,r=0.9999.The average recovery was 99.96%,RSD=1.1%.Conclusion:The reaction of chitosan with ninhydrin is sensitive and reproduc- ible.In addition,this method is simple,rapid and suitable for routine analysis of chitosan samples.
