摘  要:目的:建立1种快速、准确地测定马尿酸的方法。方法:采用 RP-HPLC 法,使用 Diamonsil^(TM) C_(18)柱(250mm×4.6mm,5μm),以乙腈-0.2%磷酸溶液(30∶70)为流动相,流速:1.0mL·min^(-1),在228nm 波长下检测血管紧张素转换酶(angioten-sin-I converting enzyme,ACE)反应中释放出来的马尿酸的量。结果:该法线性范围为1.0-5.0μg(r=0.9999),0.1-0.5μg(r=0.9998)和0.01-0.04μg(r=0.9967)。结论:该法是一种新的、快速、准确、实用的检测 ACE 抑制剂活性的方法。Objective:To establish an RP-HPLC method for estimating the angiotensin-Ⅰconverting enzyme (ACE)inhibitory activity by separating the hippuric acid liberated in the ACE reaction mixture.Method:An opti- mal RP-HPLC method was set up.The sample were separated from ACE reaction mixture was estimated by Dikma Diamonsil^(TM) C_(18) column(250mm×4.6mm,5μm)with acetonitrile-0.2% phosphoric acid(30:70)as mobile phase and detected with UV 228 nm and a flow rate of 1.0 mL·min^(-1).Result:The calibration curve was linear in the range of 1.0-5.0 μg(r=0.9999)for determination of its related substances with high concentration,0.1- 0.5μg(r=0.9998)for substance with average concentration and 0.01-0.04μg(r=0.9967)for substance with low concentration.Conclution:This is a novel,prompt,concise and useful method for estimating the angiotensin-Ⅰ converting enzyme inhibitory activity.
