摘 要:目的:观察采用化学萃取法和反复冻融法制备大鼠坐骨神经去细胞基膜管的组织形态学变化。方法:雌性Sprague-Dawley鼠坐骨神经16条随机分为化学萃取法(chemicalextractednerve,CEN)和冻融法(freeze-thawednerve,FTN)两组,每组18条。分别予Trinton-100化学萃取、-196℃~20℃反复冻融处理后,光镜、电镜观察其组织、形态学变化。结果:(1)CEN组基膜管管壁完整、贯通,无细胞结构,可见管状神经轮廓。(2)FTN组基膜管管壁破损、管腔堵塞,轴突、髓鞘、雪旺细胞及小血管变性、坏死,可见大量细胞碎片。结论:两种方法均能保持周围神经正常的三维空间结构,CEN制备的周围神经去细胞基膜管更具有组织形态学优势。Objective:To observe the morphological changes of acellular basal lamina tubes pretreated respectively by chemical extraction procedure and freeze-thawed procedure.Methods:Sixteen sciatic nerves of Sprague-Dawley rats were divided randomly into two groups-chemically extracted nerve group(CEN) and freeze-thawed nerve group(FTN).The nerves of CEN were treated with a solution of Triton X-100 and a solution of sodium deoxycholate at room temperature.The nerves of FTN were freezed at 196℃ and then thawed at 20℃.The structure changes of the nerves were observed morphologically,immunohistochemically and ultrastructurally.Results:(1) CEN:the cells,axons and myelin sheath were removed and empty basal lamina tubes were preserved.(2) FTN:Degenerated or necrotized cells?axons and myelin sheath blocked the basal lamina tubes.The membrane of basal lamina tubes was damaged.Conclusion:Both of the procedures can keep the three-dimensional space structure of the sciatic nerve,but the chemical extraction procedure has more morphological advomtages.
