摘  要:目的探讨γ射线对胱冬肽酶-3(Caspase-3)基因在犬胆管壁增殖平滑肌细胞凋亡中的活性影响。方法将103pd放射性支架和普通支架分别植入犬肝外胆管内,取出胆管标本,用免疫组化进行平滑肌细胞凋亡染色,并在JEM1200EX电镜下观察凋亡细胞超微结构变化;又用RTPCR检测γ射线诱导犬胆管壁凋亡的增殖平滑肌细胞中Caspase-3基因的mRNA。结果103pd支架组犬胆管组织中Caspase-3基因表达较普通支架组明显,且Caspase-3基因高表达组犬胆管出现明显增殖平滑肌细胞凋亡,而且犬肝外胆管无明显狭窄;而普通支架Caspase-3基因低表达,且犬胆管未出现增殖平滑肌细胞明显凋亡,而且犬肝外胆管有明显狭窄。结论辐射可以使Caspase3基因活性增高,切与细胞凋亡有关;103pd放射性支架通过激活Caspase-3基因,促进犬胆管增殖平滑肌细胞凋亡,可以抑制犬肝外胆管狭窄。Objective To research the Caspase-3 gene activity in γ-radiation-induced dog biliary duct mucle cell lines apoptosis.Methods Stenosis models of biliary duct after biliary duct intimal injury were established in dogs.The dogs were randomly divided into the ~ 103pd radioactive stent group and the control group.γ-radiation-induced apoptotic mucle cells was analysed by using immunohistochemical technique and their ultrastructural changes were observed under JEM-1200EX electron microscopy.The expression of Caspase-3 mRNA in the control andγ-radiation-induced apoptotic mucle cells was detected by RT-PCR.Resutls The expression of Caspase-3 mRNA was higher in ~ 103pd radioactive stent dog bile duct than in general stent dog bile duct,and apoptotic mucle cells were significantly increased in ~ 103pd radioactive stent dog bile duct than in general stent dog bile duct.Conclusion The high expression of Caspase-3 gene may aid the effects to understand the mucle cells sensitivity toγ-radiation apoptosis.~ 103pd radioactive stent may increase the expression of Caspase-3 gene in dog bile duct,and prevent the biliary narrow after dog bile duct was injured by balloon.
