摘 要:目的:了解不同浓度的基底膜成分对体外培养雪旺细胞生长的影响。方法:用不同浓度的基底膜成分修饰培养板底,并设置空白对照,将纯化的雪旺氏细胞以相同数量接种至各组孔内;观察细胞生长,培养72h后以MTT法和3H-TdR掺入法检测各组细胞的增殖情况,数据进行统计学分析。结果:层粘连蛋白、纤维粘连蛋白和IV型胶原促雪旺细胞分裂增殖的最低浓度分别是:10μg/ml、10μg/ml、3μg/ml;发挥最大促进效应的浓度分别是:500 μg/ml、300 μg/ml和100μg/ml。结论:不同的基底膜组份和修饰浓度对雪旺细胞分裂增殖所发挥的作用强度不同,本实验为组织工程化神经桥接体的优化修饰提供了实验基础。Objective:To observe the effects of different concentrations of basement membrane components on the growth of Schwann cell in vitro. Methods:The plates were divided into different groups and treated with basement membrane components at different concentration respectively, and then purified Schwann cells of same amount were planted into each group as above. The growth of cells was observed under inverted microscope. After 72 hours of culturing, the proliferation of Schwann cell was evaluated by MTT and -thymine nucleoside and the data was statistically analyzed. Results:The minimum concentrations of LN, FN and Col.IV that can stimulate the proliferation of Schwann cell were 10μg/ml, 10 μg/ml, 3 μg/ml and the optimal concentrations were 500 μg/ml, 300 μg/ml, 100 μg/ml separately. Conclusions:Different components and concentration of basement membrane have different effects on the proliferation of Schwann cell. Our study provides the experimental foundation for the optimal treatment of tissue-engineering bioartificial nerve.
