摘 要:[目的]研究纳米水镁石纤维(nanometerbrucitefiber,NMBF)的细胞毒性及经活性剂处理后其细胞毒作用的变化。[方法]收集大鼠肺泡巨噬细胞,培养后分别加入纳米水镁石纤维、普通水镁石纤维(commonbrucitefiber,CBF)、经活性剂处理后的纳米水镁石纤维(activator-treatednanometerbrucitefiber,ANMBF)、经活性剂处理后的普通水镁石纤维(ac-tivator-treatedcommonbrucitefiber,ACBF)、石英及生理盐水,测定肺泡巨噬细胞死亡率、乳酸脱氢酶(LDH)活力和酸性磷酸酶(ACP)活力。[结果]纳米水镁石纤维组的细胞死亡率、LDH、ACP活力分别为(37.60±8.48)%、(158.93±32.25)U/L、(25.81±10.18)U/L,均高于阴性对照组(P<0.01)和普通水镁石纤维组(P<0.05);以上指标均低于石英组(P<0.01);与经活性剂处理后的纳米水镁石纤维组相比,以上指标均有明显的升高(P<0.05)。[结论]纳米水镁石纤维具有明显的细胞毒性,且其细胞毒作用较普通水镁石纤维强,但明显要弱于石英;经活性剂处理后,其细胞毒作用明显减弱。[Objective] To study the cytotoxicity of nanometer brucite fiber and the changes of it after activator-treated. [Methods] The alveolar macrophage of rat lungs were collected, cultured, exposed to nanometer brucite fiber, common brucite fiber, activator-treated nanometer brucite fiber,activator-treated common brucite fiber,quartz dust and 0.9%NaCl and their mortality,activities of lactic dehydrogenase(LDH) and acidic phosphatase(ACP) were determined. [Results] The mortality, activities of LDH and ACP of group exposed to nanometer brucite fiber were(37.6±8.5)% ,(158.9±32.3)U/L,and(25.8±10.2) U/L,respectively. Compared with negative control group, those indicators of it increased more significantly(P<0.01); Compared with common brucite fiber, those indicators raised obviously(P<0.05); Compared with quartz dust,those indicators decreased more significantly(P<0.01); Compared with activator-treated nanometer brucite fiber, those indicators raised obviously(P<0.05) . [Conclusion] The cytotoxicity of nanometer brucite fiber was very obviously and stronger than common brucite fiber,but more weaker than quartz dust; this effect decreased obviously after activator-treated.
