四种中药复方对急性心理应激大鼠IL-1β和IL-2的影响

摘　　要：目的通过观察理气、化痰、补气、温阳方药对急性心理应激大鼠白细胞介素1β(IL1β)、白细胞介素2(IL2)的影响,研究中药复方对急性应激免疫内分泌的调节作用。方法将大鼠分为7组:正常组、模型Ⅰ组、模型Ⅱ组、四逆散组、半夏厚朴汤组、甘麦大枣汤组、桂枝甘草龙骨牡蛎汤组;观察各组大鼠的胸腺指数、脾指数及肾上腺指数,采用放免法测定各组大鼠血清IL1β、IL2的含量。结果急性心理应激后大鼠血清IL1β升高,IL2降低,胸腺指数、脾指数降低(P<0.01),肾上腺指数无变化(P>0.05);四逆散升高胸腺指数、脾指数(P<0.01、P<0.05),降低IL1β(P<0.01);半夏厚朴汤降低IL1β(P<0.01、P<0.05),升高IL2(P<0.05);甘麦大枣汤升高脾指数,降低IL1β(P<0.05);桂枝甘草龙骨牡蛎汤升高胸腺指数(P<0.01),降低IL1β的含量(P<0.01),升高IL2的含量(P<0.01)。结论四种中药复方通过作用于应激大鼠细胞因子及下丘脑-垂体-肾上腺轴的不同环节,调节急性心理应激后大鼠的免疫内分泌。Objective In order to study the regulating effects of traditional Chinese herbal compound formulae on the immuno-endocrine network of psychological stress rats, four kinds of traditional Chinese herbal compound formulae, respectively acting on regulating qi, dispersing phlegm, tonifying qi and warming Yang, were used to treat the acute psychological stress rats, and the serum contents of IL-1βand IL-2 were assayed. Methods Rats were divided into 7 groups: normal group, model 1 and 2 groups, Sini Powder group, Banxiahoupu Decoction group, Ganmaidazao Decoction group, and Guizhigancaolonggumuli Decoction group. The indexes of thymus gland, spleen and adrenal gland were observed; the serum contents of IL-1βand IL-2 of all the rats were detected by the means of radioimmunoassay. Results In acute psychological stress rats,the serum content of IL-1β was higher, but the level of IL-2, the indexes of thymus gland and the spleen were lower (P<0.01), while the index of adrenal gland had not any obvious change (P>0.05); Sini Powder increased the indexes of thymus gland and the spleen (P<0.01, P<0.05), and decreased content of serum IL-1β(P<0.01); Banxiahoupu Decoction decreased serum content of IL-1β(P<0.01, P<0.05) and increased the content of IL-2 (P<0.05); Ganmaidazao Decoction increased the index of the spleen, and decreased the content of IL-1β(P<0.05); Guizhigancaolonggumuli Decoction increased the index of the thymus gland and the content of IL-2 (P<0.01),and decreased the content of IL-1β(P<0.01). Conclusion The four kinds of traditional Chinese herbal compound formulae can regulate immuno-endocrinel network of the acute psychological stress rats through the action on the cytokine and the different taches of the hypothalamus-pituitary-adrenal gland axise.
