摘  要:目的探讨L卡尼汀对大鼠心脏移植再灌注损伤心肌热休克蛋白70(HSP70)表达的影响。方法采用大鼠颈部异位心脏移植模型,将SD大鼠随机分为2组:L卡尼汀组和对照组。采用免疫组化和Westernblot检测再灌注2h心肌HSP70的表达,电镜观察心肌线粒体的病理变化。结果L卡尼汀组心肌HSP70的表达较对照组明显升高(P<0.01),心肌线粒体等结构损伤明显减轻。结论L卡尼汀对移植再灌注损伤心肌的保护作用可能与其诱导心肌HSP70的表达有关。Objective To explore whether L-carnitine has an effect on the expression of myocardium heat shock protein 70 (HSP70) in rat heart transplantation reperfusion injury.Methods The model of rat cervical heterotopic heart transplantation was set up. L-carnitine was injected to the recipient rats 4 h before transplantation. The expression level of HSP70 protein was measured by immunohistochemistry and Western blot after 2 h. Pathological changes of myocardia especially mitochondria was observed under transmission electron electronmicroscopy.Results The expression of HSP70 was significantly higher in L-carnitine group than that in control group (P<0.01). The lesion of myocardia ultrastructure especially mitochondria in L-carnitine group was significantly milder than in control group.Conclusion L-carnitine can increase the expression of HSP70 in rat heart transplantation and alleviate the pathological lesions of mitochondria. L-carnitine may have some protective effects on the reperfusion injury in rat heart transplantation.
