摘　　要：目的离子对-反相高效液相色谱(HPLC)法测定人血清中美托洛尔浓度的评价。方法血清样品经蛋白沉淀剂(N,N-二甲基甲酰胺)沉淀后,高速离心,取上清液进行分析。测定使用C18色谱柱,甲醇-水(50:50,水相中含0.14%磷酸二氢钾)为流动相,以VP-16为内标,荧光检测Ex=277nm,Em=299nm。结果美托洛尔的浓度检测范围是2～1000pg/ml,r=0.9988;样品平均回收率为94.15%,相对标准差RSD为3.62%(n=6)。结论本方法选择性强,灵敏,重现性好。Objective To establish a method for the determination of metoprolol in serum by IP-RP HPLC (ion-pair RP High performance liquid chromatography).Methods The extracts of samples were dopesited by N,N-Dimethyformamide,centrifuged with high speed,then the liquid of superstratum was taked to analyze on a C18 column with methanol-aquous solwtion(0.14%)(50:50) VP-16 was internal standard.The dromtogroph was monitored by fluorescence detection(Ex=277 nm,Em=299 nm).Results The ranger of metoprolol detected was from 2 to 1000 pg/mL and correlation coefficient was 0.9988The average recovery of sample was 94.15% and RSD=3.62%(n=6),respectively.Conclusion The HPLC is a sensitive method with good selectivity and reproducibility.
