摘 要:研究外植体的大小和灭菌方式对柑橘茎段外植体消毒效果的影响·结果表明,0.5cm长的柑橘纵切茎段外植体污染率较低;用70%的酒精处理30s,10%的次氯酸钠溶液处理30min,放置48h后,再用10%的次氯酸钠溶液处理30min,成功率最高;高温干燥季节取材,污染率较低;不同品种的污染率和发芽率有一定差异.The effect of explant size and disinfection protocols on the disinfection of citrus stems with lateral buds has been tested. The result showed that the contamination rate of longitudinally-cut inter-node explant of citrus about 0.5 cm long was lower. After treated by 70% ethanol for 30s and followed by 10% hypochlorous sodium for 30 min, the materials were kept in culture room for 48 h, then the material was treated by 10% hypochlorous sodium for 30 min again. By this method, the success rate was the highest; the contamination rate of the materials took in hot and dry season was lower; Difference of contamination and sprout rate existed among cultivars .
