Detection of Hepatitis D Virus RNA in Liver Tissue by in situ Hybridization with Digoxigenin-labeled HDV cDNA probes

摘  要:本研究采用随机引物法用地高辛标记HDV cDNA全基因片段作为探针,通过原位杂交方法检测了58例慢性乙型肝炎患者的肝组织石蜡切片中的HDV RNA,发现HDV RNA阳性者9例,检出率为15.5%。HDV RNA阳性物质在肝细胞内的分布方式有胞浆型、核膜型、核仁型、核膜核仁型和全核型等。HDV RNA阳性肝细胞在显微镜下多数正常或只显示轻微病变。本文结果提示HDV的致病性并非由于HDV的直接细胞毒作用,而是宿主细胞和病毒共同作用结果,但其确切生物学意义还有待进一步探索。Whole gene HDV cDNA labeled with digoxigenin through random primer DNA labeling method was used to detect HDV RNA in formalin-fixed paraffin-embedded section by in situ hybridization. In nine of fifty eight patients with chronic hepatitis B (15.5%),HDV RNA was found in liver tissue and was distributed in cytoplasmic,nuclear membrans,nucleolar,nuclear membrane and nucleolar,and whole nucleus pattern. No morphological change or only minimal change was observed in those hepatocytes with HDV RNA. The results suggest that the cytopathic effect of HDV on hepatocytes is not caused by direct cytotoxicity and may be,however,due to interaction of host cells and virus.
