2型糖尿病人群胰岛素抵抗和β_3-AR基因的相关性

摘　　要：目的探讨β3肾上腺素能受体基因与2型糖尿病胰岛素抵抗的关系。方法对131例糖尿病患者和40例正常对照者,应用PCR-RFLP技术扩增β3-AR基因,抽血测定空腹血糖、总胆固醇、甘油三酯、高密度脂蛋白胆固醇,糖化血红蛋白、胰岛素。结果2型糖尿病与正常人群β3-AR基因型和等位基因频率没有差异,但在2型糖尿病组,携带β3-AR基因64位Arg突变的患者,其胰岛素抵抗指数(HOMA-IR)显著高于无突变组(P<0.05)。结论β3-AR基因多态与2型糖尿病胰岛素抵抗有相关性。Objective To explore the association between β_3-adrenergic receptor gene polymorphism and insulin resistance in type 2 diabetics.Methods Fast serum levels of glucose,blood lipids and insulin and HbA_1c were detected in a population of 131 type 2 diabetic patients and 40 non-diabetic subjects. All subjects were genotyped for the β_3-adrenergic receptor gene polymorphism by using PCR/BstNI restriction endonudease digestion.Results The distribution of β_3-AR gene polymorphism between type 2 diabetes and controls is not different.the HOMA-IR value of those with β_3-adrenergic receptor gene Trp/Arg mutation were significant higher than that of those without the mutation in type 2 diabetes.Conclusion This suggests that there is an association between the Trp/Arg mutation of the β_3- adrenergic receptor gene and insulin resistance in Chinese with diabetes mellitus.
