摘 要:目的 观察表皮细胞生长因子 (EGF)在体内是否具有诱导骨髓间充质干细胞 (MSCs)向皮肤组织分化的作用 ,及其在皮肤创面愈合中的作用。方法 抽取小型猪骨髓 ,体外分离、纯化和培养MSCs ,并用5 溴脱氧尿嘧啶 (BrdU)进行标记。 6只小型猪共 4 8个创面 ,随机分为EGF +MSCs治疗组 (A组 )、MSCs治疗组 (B组 )、EGF治疗组 (C组 )和生理盐水对照组 (D组 )。观察伤后治疗 3、7、14、2 1及 84天的创面 ,并用病理学、免疫组化方法对创面的修复质量进行评估。结果 A组的皮肤创面愈合速度明显快于其它治疗组 ,并且在皮肤愈合的组织病理等级评分上优于B、C和D组。A组表皮层BrdU和CKp双染阳性细胞比B组更为多见。结论 EGF具有在体促进猪骨髓间充质干细胞向表皮细胞分化 ,加快皮肤创面愈合速度和提高愈合质量的作用。Objective To investigate the role of epidermal growth factor (EGF) on the differentiation of mesenchymal stem cells to skin tissues in vivo and its effect on skin wound healing.Methods MSCs was isolated from porcine marrow,then purified and cultured with 5-bromodeoxyuridine(5--BrdU) as a label in vitro.Forty-eight skin wounds on the 6 pigs were divided into EGF+MSCs (A),MSCs(B),EGF(C) and contrast (D) groups randomly and treated correspondingly.Wound closing index (WCI),cell morphology and histological changes in the defected regions were evaluated 3、7、14、21、84 days after treatment.Results Curative speed of defected regions of group A was faster than that of the other groups;Hierarchical scores of histopathology on the specimens of group A was also higher than that of the other groups.The dual-positive stained cells by BrdU and CKp were more in the specimen of the group A than that of group B.Conclusion EGF can induce MSCs to differentiate into epidermal cells,and promote the repair of skin wounds as well.
