摘  要:目的:研究阳离子脂质体介导体内外基因转移的效率。方法:采用逆相蒸发法制备出包裹TNFαDNA的脂质体,将其与体外培养的靶细胞共育,或将其直接注射至小鼠腹腔内,检测TNFα的分泌水平。结果:TNFα基因转染NIH3T3细胞和腹腔细胞,10d后均可检测到TNFα的分泌。结论:研制的脂质体不但能将目的基因转移至体外培养的靶细胞中,还可通过腹腔内注射直接将TNFα基因转移至腹腔细胞,稳定表达TNFα。Objective:To investigate the efficiency of cationic liposome in mediating gene transfer. Methods: With reverse-phase evaporation, we prepared cationic liposome encapsulated TNFα DNA, which were either co-cultured with those in vitro cultured target cells, or were directly injected into the abdominal cavity of mice. Then, TNFα secretion level was measured. Results: TNFα secretion could be detected in the NIH3T3 cells and the peritoneal cells transfected with TNFα genes, 10 days later. Conclusion: Results show that liposome can mediate cytokine gene transfer efficiently in vitro and in vivo, and TNFα can be expressed stably.
