摘 要:用免疫组织化学ABC法检测了SD大鼠胚胎(E13、14、18、20d)和幼鼠(P1、3、6d;2周、3周)体内运动神经元诱向因子(MNTF)的分布。结果表明,大鼠E18d之前的胚胎组织或脏器的MNTF-McAb免疫组织化学反应均为阴性。在大鼠E18d的心肌、骨骼肌呈免疫阳性反应,而平滑肌为阴性反应;肝、肾、脊髓的边缘层和套层的原始神经细胞、胶质细胞及E20d前角运动细胞均为阳性反应。从P1d到3周龄大鼠的心肌、骨骼肌仍为阳性反应;肝和肾的阳性反应逐渐减弱;胰腺泡上皮细胞为阴性反应,但胰岛内分泌细胞免疫阳性反应明显,而且随着动物成长其阳性反应逐渐增强,直至成体大鼠的胰岛才转为阴性反应。在3周龄动物,主要是脊髓前角运动神经元有明显的阳性反应。实验结果表明,MNTF在SD大鼠胚胎晚期才出现,而且分布范围比较局限。提示MNTF在体内的主要功能为促进其效应细胞(运动神经元)的生长、成熟和行使功能。In this paper monoclonal antibody specific to motoneuronotrophic factor was used to study the immunohistochemical distribution of the motoneuronotrophic factor in embryos(E13,14,18and 21 days)and neonates(P1,3,6,14 and 21 days)of SD rats.The results revealed that viscera and tissues from embryos younger than 18 days were immunoreactively negative;those from embryos 18 days onward including cardiac and skeletal muscles, liver and kidney,and primitive neurocytes and neuroglia present in the marginal and mantal layers of the spinal cord,as well as the anterior horn motoneurons in the E20 spinal cord were immunoreactively positive;but not the smooth muscles.For neonatal rats from 1 to 21 days old,their cardiac and skeletal muscles were still immunoreactively positive and their liver and kidney were gradually becoming weakly positive.Their pancreatic acinar cells were negatively stained,however,the pancreatic islet cells were distinctively becoming strong immunoreactively positive until in adult rat,and then became immunoreactively negative.The spinal cord was weakly immunoreactive positive in newborn rats but the immunoreactivity in its anterior horn motoneurons became distinctively strong positive in 21 day old rats.These results indicated that the motoneuronotrophic factor began to be expressed quite locally during the late embryonic period and later more concentrated in the neuromuscular system,and suggested that the major role of the motoneuronotrophic factor is mainly involved in promoting the growth,maturation and function of the motoneurons.
