人眼虹膜色素上皮细胞体外培养及超微结构观察

摘　　要：应用体外培养传代对１０例人尸眼虹膜色素上皮细胞进行观察分析。结果表明：直接刮取的细胞进行培养较酶消化法更易存活传代。原代细胞在光镜下呈多角形单层生长，胞浆内有丰富的色素颗粒，核相对透明，有１～２个核仁。传代后的细胞色素颗粒减少；电镜下见胞浆富含色素，细胞膜有明显微绒毛；免疫组化检查角蛋白抗原呈阳性反应，符合虹膜色素上皮细胞的特点。虹膜含有某些生物活性物质，因此虹膜色素上皮细胞的体外培养可作为有关研究的基础。AbstractThe iris pigment epithdial cells of 10 human eyeswere cultivated in vitro. Results:The iris pigment ep-ithelial cells obtained directly by scraping were easierto grow and to be passaged than those obtained by en-zyme digestion. Under the inverted light microscope,primary cells appeared multigonal and arranged inmonolayer, there were abundant pigment granules inthe cytoplasm, and the nuclei each of which contained1 or 2 nuceoli were relatively transparent. Pigmentgranules diminished in succeeding generations; Underthe transmission electron microscope, pigment gran-ules were rich in the cytoplasm and there were plentyof microvilli at teh cell membrane。 There were macu-lae occludentes and desmosomes present in the intercellular space。 The cultivated cells showed positive re-action of keratin antigen in an immunohistochemicalassay. The characteristics mentioned above are consis-tent with the criteria of iris pigment epithelial cells.
