摘 要:目的:研究赤芍总苷(totalsaponinsofpaeonia,TSP)和淫羊藿总黄酮(totalflavonesofepimediumgradiflorummaxim,TFE)及其不同比例复合物对实验性抑郁的影响,并探讨长期给药对慢性应激合并不完全脑缺血再灌注抑郁大鼠大脑5-HT和β-肾上腺素受体密度的影响。方法:通过小鼠悬尾法、游泳绝望法研究受试药物对实验性抑郁行为的影响;通过小鼠5-HTP增强实验和小鼠NA毒性增强实验研究药物的作用途径;通过慢性随机刺激合并结扎/开放双侧颈总动脉制备抑郁模型,并测定其海马组织β-肾上腺素受体和大脑皮层5羟色胺受体密度,研究长期给药对神经递质受体的影响。结果:TSP、TFE及其不同比例复合物均能不同程度改善实验性抑郁行为,且与单胺类递质有一定关系。慢性随机刺激合并结扎/开放双侧颈总动脉可造成大鼠海马组织β-肾上腺素受体和大脑皮层5-HT受体密度上调,给予TSP及TSP/TFE复合物能将其调低,而TFE仅对β-肾上腺素受体产生影响。结论:TSP、TFE均有一定抗抑郁作用,且TSP作用可能更为广泛;TSP/TFE不同比例复合物作用类似,优于单独给予TSP或TFE,且以2∶1时作用最优;TSP和TFE治疗血管性抑郁的机制可能较为广泛,至少与NA、5-HT等单胺类神经递质及其脑内不同区域受体有关。Objective:To research the effect on experimental depression of TSP/TFE and their mixture of different proportion, and discuss the effect on 5-HT in brain and the density of β-adrenergic receptor by a long-term administration on chronically stimulated and Semi-cerebral ischemia re-perfuse depression rat.Methods:The effect on experimental depression were researched by mice tailing hanging and swimming;the acting approach were researched by enhancing 5-HTP and NA toxicity of mice;the depression model was set up by chronic stimulation and ligate/perfuse double carotid, and the density of β-adrenergic receptor in hippocampus and 5-HTP receptor in cerebral cortex was determined,and the effect on neurotransmitter of a long-term administration was discused.Results:TSP/TFE and their mixture of different proportion could improve experimental depression behavior at different degree, relating with monoamine transmitter. And chronic stimulation and ligating /perfusing double carotid could increase the density of β-adrenergic receptor in hippocampus and 5-HT receptor in cerebral cortex. Administration of TSP and TSP/TFE compound could decrease the density and TFE only affect on β-adrenergic receptor. Conclusion:Both TSP and TFE have some anti-depress function and TSP may have wider range; their mixture of different proportion have the similar function but better than single either, and the most effective ratio is 2:1; the mechanism of TSP and TFE treating vascular depression may be comprehensive, except for the relationship with NA ,5-HT and similar monoamine transmitters and receptors of different areas in brain.
